## Indications for Use

Cryo✓Check™ PNP Platelet Lysate is intended for use in the Platelet Neutralization Procedure (PNP). The PNP is useful in detecting the presence of lupus anticoagulants in human plasma.

## Device Story

Cryo✓Check™ PNP Platelet Lysate is a reagent derived from human platelets collected from healthy donors, washed, resuspended in buffer, and processed via freeze-thaw cycles to create a suspension of ruptured platelet membranes. The device is used in clinical laboratories as part of the Platelet Neutralization Procedure (PNP) to detect lupus anticoagulants in human plasma. The reagent is aliquoted into cryovials and stored frozen. During testing, the lysate is mixed with patient plasma; the presence of lupus anticoagulants is identified by comparing the Activated Partial Thromboplastin Time (APTT) of the plasma-lysate mixture against a saline-plasma control. The resulting correction in clotting time assists clinicians in diagnosing the presence of lupus anticoagulants, which are associated with certain clinical conditions. The device is compatible with automated coagulation analyzers using either photo-optical or mechanical clot detection systems.

## Clinical Evidence

Bench testing only. Performance was evaluated by comparing PNP results of normal and known lupus anticoagulant-positive patient samples using the subject device versus the predicate. Stability studies included an 8-hour open-vial study and a 14-hour on-board stability study on an MDA-180 analyzer. Consistency was verified across two different automated coagulation systems (photo-optical and mechanical). Results showed comparable specificity to the predicate and consistent vial-to-vial reactivity.

## Technological Characteristics

Reagent consists of ruptured human platelet membranes in buffer. Supplied as a frozen liquid in cryovials. Compatible with automated coagulation analyzers (e.g., MDA-180, Stago ST4). Sensing principle relies on standard coagulation clot detection (photo-optical or mechanical) following the addition of the platelet lysate reagent to patient plasma.

## Regulatory Identification

A partial thromboplastin time test is a device used for primary screening for coagulation abnormalities, for evaluation of the effect of therapy on procoagulant disorders, and as an assay for coagulation factor deficiencies of the intrinsic coagulation pathway.

We have reviewed your Section 510(k) notification of intent to market the device referenced above and we have determined the device is substantially equivalent (for the indications for use stated in the enclosure) to legally marketed predicate devices marketed in interstate commerce prior to May 28, 1976, the enactment date of the Medical Device Amendments, or to devices that have been reclassified in accordance with the provisions of the Federal Food, Drug, and Cosmetic Act (Act). You may, therefore, market the device, subject to the general controls provisions of the Act. If your device is classified (see above) into either class II (Special Controls) or class III (Premarket Approval), it may be subject to such additional controls.
