Intended Use

The VisuLize™ Factor VIII Antigen kit is an Enzyme Immunoassay for the quantitative determination of Factor VIII antigen in human plasma samples and Factor VIII concentrates using the double antibody enzyme linked immuno-sorbent assay (ELISA).

Device Story

VisuLize™ Factor VIII Antigen Kit is an ELISA-based in vitro diagnostic assay; utilizes pre-coated 96-well plates with sheep polyclonal antibody to capture Factor VIII from diluted human plasma or concentrates. Process involves: sample application; washing unbound proteins; adding horseradish peroxidase (HRP)-labeled polyclonal detection antibody; adding TMB chromogenic substrate. Color development measured spectrophotometrically at 450 nm; absorbance directly proportional to Factor VIII concentration. Used in clinical laboratory settings by trained personnel. Output provides quantitative Factor VIII antigen levels, aiding in the assessment of Factor VIII deficiency or monitoring. Benefits include standardized, sensitive quantification of Factor VIII antigen compared to activity-based assays.

Clinical Evidence

Clinical performance compared via testing of 323 total samples across three sites (internal and two external). Results showed high correlation with the predicate device: Pearson correlation coefficients (r) were 0.968 (internal), 0.964 (site 1), and 0.974 (site 2).

Technological Characteristics

Sandwich ELISA; 96-well format pre-coated with sheep polyclonal antibody. Detection via HRP-labeled polyclonal antibody and TMB substrate. Spectrophotometric measurement at 450 nm. Calibrated against WHO International Standard (97/586). Requires 3.2% sodium citrate plasma. Bench testing performed per CLSI guidelines (EP5-A, EP17-P, EP7-P).

Indications for Use

Indicated for the quantitative determination of Factor VIII antigen in human plasma samples and Factor VIII concentrates via ELISA. Intended for in vitro diagnostic use.
